It is recommended that a minimum concentration of 250-500 ng/µL of amplified DNA be used for spotting each sample. Synthetic oligonucleotides (10-100 picomole/µL) made from expressed sequences (ESTs) can be also used. For target preparation and hybridization, either 100 µg of total cell RNA or 5 µg polyA+mRNA is required in 1-15 µL volume. Typically, a single clone is used to analyze each mRNA.
